Title:  Neurobiology Interest Series: Structural basis of fatty acyl recognition and transfer by the DHHC20 palmitoyltransferase enzyme
Description:  DHHC palmitoyltransferases are a family of integral membrane proteins that catalyze the S-acylation or palmitoylation of protein substrates. Since their discovery in early 2000s, DHHC enzymes have generated considerable interest as palmitoylation plays critical roles in biology such as meiosis, parasite development, immunity, and plant development among others. The brain is a particularly active area of palmitoylation and this is known to play important roles in neuronal development and synaptic plasticity. Dysregulation of protein palmitoylation has been linked to human diseases such as cancer and nervous system disorders. Despite the general importance and biomedical relevance, mechanistic understanding of DHHC enzymes has lagged due to the lack of any high-resolution structure. Here we present the ~ 2.5-Å structure of the human DHHC20 enzyme. Combined with mutational analyses, the structure gives insights into the overall organization of the DHHC enzyme and the chemical basis of catalysis. In particular, we clarify the roles of the TTxE and PaCCT conserved motifs of the DHHC palmitoyltransferases. We also present a covalently inhibited structure of the DHHC enzyme that represents an intermediate in the catalytic cycle. This structure unambiguously allows us to define the fatty acyl chain binding groove in the protein and provides a stereochemical basis for fatty acyl selectivity.
