Title:  VIG Seminar: The Art of War: Exosomes as Carrier Pigeons of the Cell to Help Protect against Pathogenic Infections
 
Description:  Exosomes are a subclass of extracellular vesicles (EVs) that have garnered huge interest in recent years as their central role in cell-cell communication and their critical impact on progression of a number of different human diseases has been discovered, including cancer and infectious diseases. However, our understanding of the molecular mechanisms by which exosomes carry out their functions is very much at its infancy. Our laboratory has been pioneering in studies of exosomal functions during infection with highly pathogenic agents such as Yersinia pestis (Yp), the agent of plague, and the Rift Valley fever virus (RVFV), the causative agent of the Rift Valley fever (RVF) disease. We have demonstrated that exosomes released from RVFV-infected cells (EXi) contain viral RNA (L, M, and S segments) and also the viral nucleocapsid protein (N) and envelope glycoproteins. Interestingly, EXi treatment of recipient immune cells, such as monocytic cells that serve as reservoirs of viral replication, blocks their ability to proliferate. In addition, pre-treatment of human HSAEC cells with EXi before infection with RVFV significantly reduces virion production and release. We have also demonstrated that IFN-ß, a target of inhibition by RVFV, is induced in these cells by EXi treatment, providing a mechanism by which EXi may be inhibiting virus production. Our results present a model in which EXi provides a protective role for the host against RVFV infection, and suggest the potential of using EXi as natural vaccines.
