Title:  Computational Modeling and Immunogen Design for a Hepatitis C Virus Vaccine
Description:  Hepatitis C virus (HCV) is a major global health concern, and despite the development of effective antiviral therapies, a preventative vaccine for HCV is a major unmet need. Due to the high sequence variability of HCV, an effective immune response must target key conserved epitopes of the virus to prevent escape. Based on the x-ray structure of a broadly neutralizing antibody in complex with a conserved epitope of the E2 envelope glycoprotein, we designed several immunogens to elicit responses to this site, leading to neutralizing antibodies in vivo. Recent work has produced designs based on other E2 epitope structures, which show the ability to selectively engage a key class of monoclonal antibody. We are also utilizing computational approaches to model E2 and its antibody and co-receptor recognition, based in part on analysis of large experimentally determined global alanine scanning data measured using a panel of monoclonal antibodies.
